Host factors in hepatitis B virus cccDNA formation as novel antiviral targets and biomarkers - identification, preclinical evaluation and impact for liver disease. - Chronic hepatitis B (CHB), caused by hepatitis B virus (HBV), is a leading cause of liver cirrhosis, liver failure and hepatocellular carcinoma (HCC). Of the 350 million chronic HBV carriers worldwide ~20% will die from terminal liver disease. 14 million Europeans live with CHB; the associated mortality rate is ~36,000 deaths/year. Current treatments offer at best control of infection but no cure. Chronicity relies on a unique intracellular viral replication intermediate termed covalently closed circular (ccc) DNA which ensures progeny virus production; its eradication is the prerequisite for a cure. However, how cccDNA is generated from the relaxed circular (RC) DNA genome present in infectious virions is not known, except that the multiple conversion steps required must largely depend on host factors. To identify such host factors as novel antiviral targets, this proposal combines the complementary competences of four HBV-experienced partner labs (Baumert & Brino, France; Bielawski, Poland; Nassal, Germany)

Precision cut lung slices: a novel versatile tool to examine host–pathogen interaction in the chicken lung - AbstractThe avian respiratory tract is a common entry route for many pathogens and an important delivery route for vaccination in the poultry industry. Immune responses in the avian lung have mostly been studied in vivo due to the lack of robust, relevant in vitro and ex vivo models mimicking the microenvironment. Precision-cut lung slices (PCLS) have the major advantages of maintaining the 3-dimensional architecture of the lung and includes heterogeneous cell populations. PCLS have been obtained from a number of mammalian species and from chicken embryos. However, as the embryonic lung is physiologically undifferentiated and immunologically immature, it is less suitable to examine complex host–pathogen interactions including antimicrobial responses. Here we prepared PCLS from immunologically mature chicken lungs, tested different culture conditions, and found that serum supplementation has a detrimental effect on the quality of PCLS. Photoluminescence of Eu2+-Doped Strontium Cyanamide: A Novel Host Lattice for Eu2+ - Eu2+-doped SrCN2 with different Eu2+ contents were prepared by a high-temperature solid-state reaction under NH3 atmosphere. SrCN2 has an energy gap of 4.56 eV by the reflection spectra. The photoluminescence spectra of SrCN2:Eu2+ at different temperatures were measured. The results show that Eu2+-doped SrCN2 shows a broad band emission with a peak at 610 nm when it is excited by 350-520 nm at 77 K. With the temperature increasing, the emission intensity decreases remarkably, indicating that Eu2+-doped SrCN2 has a low quenching temperature.
